Performance
To ensure highest sensitivity, the ipsogen JAK2 MutaSearch Kit has been optimized to detect the JAK2 V617F/G1849T mutation and uses a DNA cut-off sample with 1% (COS) positivity and DNA positive and negative controls for qualitative analysis (see figuresAccurate detection of JAK2 V617F mutation, 1% cut-off sample, and positive and negative controls and Reliable detection of wild-type JAK2, 1% cut-off sample, and positive and negative controls). A minimum of 25 ng of input sample DNA is recommended.
Principle
The ipsogen JAK2 MutaSearch Kit is a ready-to-use kit for the detection and qualitative analysis of the JAK2 V617F/G1849T somatic mutation due to a single nucleotide polymorphism (SNP) in genomic DNA (see figure JAK2 mutation site identification). The kit provides high levels of sensitivity and reproducibility. The ipsogen JAK2 MutaSearch Kit provides a DNA cut-off sample (COS), a positive control (PC), and a negative control (NC) for JAK2 V617F. Use of the cut-off sample and controls enables accurate qualitative analysis of genotyping results.
Procedure
The first step is extraction and determination of quality and quantity of genomic DNA. The second step is the amplification of DNA by real-time PCR. Using the ipsogen JAK2 MutaSearch Kit provides sensitive detection and qualitative analysis of the JAK2 V617F/G1849T mutation. Simply start the reaction using the optimized protocols described in the kit handbook.
